That's a contradiction in terms. JAMMA standard is 2 players, 3 (or 4) buttons. Some games have harnesses for extra buttons or extra controllers. You have to wire these as you go. Of course, you can make a unit that has 4 ports on it... but you'll then have to wire the extra ports / leads to harnesses. How are you going to do that when they're not standard? The answer to your question is no - it's a standard JAMMA wiring, plus you can add whatever harnesses you see fit as you wish.
The easiest way would be to have screw terminals at the back of the unit which are connected to what every control port you are going to use, then you use the loom from the 3P / 4P pins on the board (and they are different, although Konami had a fairly static pin out for this) and screw the tinned wires from the loom into the supergun, helps if the wires are easily identified.
Yeah, or a multi-pin connector on the unit and make harnesses for whatever boards you want to use it with... much like you would for a fingerboard, really.

Exactly. The extra players usually have standalone pinouts so making a separate harness for them is easier, you can even make a kick box for the 3p and 4p harnesses if you want.
