    Not knowing anything about NES protos, the picture you posted makes it very much look like a reproduction, IE, a fake. The model and serial number appears to have been filed off the exposed chip, probably to hide when it was manufactured, the tape over the EEPROM windows looks very new, and the whole board itself just seems a little too "clean". I could make something very similar looking in a few hours with what I have lying around in my study. Since Google tells me that "Escape from Atlantis" is a prototype that's already been dumped for the NES, I'd say you're going to have to provide a lot more evidence and information, IE, in particular some pictures of the game running, in particular showing any differences from the currently available ROM, and a good explanation for some of the points of suspicion, before anyone is willing to part for any sum of money on this game.
